Add a button on node view: panels or just use a block.
Add to the job-post node: I'm not sure exactly what you mean, but again, Panels or use a Views block.
I don't understand what you want with 2.

I have created a couple of roles employer and job seekers. The employer can create a content I called job-post and the job seekers can view these content. I have also included a user reference field in the job-post content that will hold the job seekers that will apply for the job-post. I am using D6. My questions would be:
1. How do I add a button on the node view of the job-post content so that when the job seekers view it they can apply and their record will be added on the job-post node?
2. After I have added the button how can I programatically add a record to the user reference field?
I looked at panels, rules and webform it seems that these modules cannot help me.
