Periodically... I'd think is throttle kicking in.
Otherwise I'd say something wrong in alias and/or admin/settings -> Default front page

I just went to your front page and it took quite a while for it to load on my machine. I'm on DSL so it shouldn't take that long. Could the page be timing out and giving the not found error?
